Ancient DNA reveals reproductive barrier despite shared Avar-period culture
Abstract After a long-distance migration, Avars with Eastern Asian ancestry arrived in Eastern Central Europe in 567 to 568 ce and encountered groups with very different European ancestry1,2. We used ancient genome-wide data of 722 individuals and fine-grained interdisciplinary analysis of large seventh- to eighth-century ce neighbouring cemeteries south of Vienna (Austria) to address the centuries-long impact of this encounter1,2. We found that even 200 years after immigration, the ancestry at one site (Leobersdorf) remained dominantly East Asian-like, whereas the other site (Mödling) shows local, European-like ancestry. These two nearby sites show little biological relatedness, despite sharing a distinctive late-Avar culture3,4. We reconstructed six-generation pedigrees at both sites including up to 450 closely related individuals, allowing per-generation demographic profiling of the communities. Despite different ancestry, these pedigrees together with large networks of distant relatedness show absence of consanguinity, patrilineal pattern with female exogamy, multiple reproductive partnerships (for example, levirate) and direct correlation of biological connectivity with archaeological markers of social status. The generation-long genetic barrier was maintained by systematically choosing partners with similar ancestry from other sites in the Avar realm. Leobersdorf had more biological connections with the Avar heartlands than with Mödling, which is instead linked to another site from the Vienna Basin with European-like ancestry. Mobility between sites was mostly due to female exogamy pointing to different marriage networks as the main driver of the maintenance of the genetic barrier.

A tale of be(com)ing a Sport Nutritionist: Reflexive insights from a researcher-practitioner entering the field
Qualitative research can facilitate an understanding of the richness and complexity of human experience, shedding light on multifaceted relationships that exist within sporting environments. Within the field of sport nutrition, such immersive methods are not commonplace. Yet could offer a significant contribution to our understanding and help practitioners navigate their values and identity within the challenging world of high-performance sport. The first author, a neophyte sport nutrition researcher-practitioner, reflects upon her experience over a four-year period of integrating into a high-performance organisation and embarking upon a research programme and career as a sport nutritionist. Drawing on field notes, reflexive journal entries, and regular research team discussions, three moments of discovery are shared: 1) Navigating the transition into high-performance sport, 2) Understanding the performance pressures and emotional burden experienced by athletes, and 3) Negotiating collaboration in the nutritionist-athlete relationship. Through this confessional tale, we recognise the development of a sport nutritionist’s values and identity is unlikely to be smooth or linear. Instead, it may be a bumpy voyage of self-inquiry and discovery, shaped by diverse cultural experiences. To help future sport nutrition researcher-practitioners entering the field, we offer three meaningful development activities including 1) engaging in reflexive practice with critical friends to explore the dilemmas and uncertainties that may impact personal and professional development, 2) fostering intellectual candour when engaging in reflexive practice to facilitate self-discovery and growth, and 3) prioritise reading the growing literature on sport and organisational culture to inform and influence the delivery of sport nutrition practice.

Enhancing nutritional value and quality of cookies through pumpkin peel and seed powder fortification
Background Recently, the pumpkin by-products such as peels and seeds become more and more interesting for confectionary sector, because of the nutritional and bioactive composition. The study was conducted to develop cookies and noodles using different pumpkin by-products (such as peels and seeds) powders in different concentrations and to evaluate the changes in quality characteristics. The powders were prepared from pumpkin seeds and peels. Methodology In this study, various concentrations of pumpkin peel powder (5%, 10%, and 20%) and seeds powder (5%, 10%, 15%, and 20%) were mixed with commercially available wheat flour to produce cookies and noodles. Furthermore, the nutritional, antioxidant, functional and sensory properties of the developed cookies and noodles were evaluated. Result Pumpkin peels and seeds powder and their respective products enrich significantly in protein, fiber, beta-carotene, antioxidants such as total phenolic content (TPC), 2,2- diphenyl-1-picrylhydrazyl (DPPH scavenging activity), Ferric reducing ability (FRAP Ferric reducing power assay), ash content and hunter color values. However, pumpkin seed powder had higher protein content (33.25%) than other powders but pumpkin peel powder (16.83%±1.74) had better fiber content than seeds powder. The highest amount of fiber content was found in 10% pumpkin peel cookies. The analysis of the composition of pumpkin peels and seeds powder has been done in order to further promote their functionality in bakery products based, especially, on wheat flour. Conclusion The best quality and overall acceptable product were obtained when samples were 10% pumpkin peel powder, 15% pumpkin seed powder, and, 10% pumpkin peel powder noodles.

Predominance of extensively-drug resistant Acinetobacter baumannii carrying bla OXA-23 in Jordanian patients admitted to the intensive care units
Background and Aim The global emergence of Acinetobacter baumannii is of great concern, especially inside intensive care units (ICUs). This study investigated the prevalence, antibiotic resistance, biofilm formation and genetic relatedness of A. baumannii recovered from ICU patients in three major hospitals in Jordan. Methods The A. baumannii isolates included in this study were identified by the detection of the blaOXA-51 gene, and a multiplex PCR assay. Antibiotic susceptibility testing was performed using the disk diffusion and broth microdilution methods, and the ability of the isolates to form biofilms was tested using the 96-well plate assay. All isolates were tested for the presence of carbapenemases-encoding genes by PCR. Clonal relatedness was assessed by Rep-PCR and dendrogram analysis. Results Overall, 148 A. baumannii isolates were identified, with 96.7% of the isolates recognized as carbapenem resistant A. baumannii. Based on their resistance patterns, 90% of the isolates were extensively resistant (XDR). The highest prevalence of carbapenemases-encoding genes was for blaOXA-23-like (96.7%), followed by blaADC (93.9.2%), blaVIM (56.8%) and blaNDM-1 (7.4%). Almost 80% of the isolates were able to form biofilms, with 63.2% classified as strong biofilm former. Rep-PCR and clustering analysis revealed 26 different clusters and the circulation of hospital-specific clones. Conclusions Our study revealed an alarming high prevalence of XDR, blaOXA-23-carrying and strong biofilm-producing A. baumannii among ICU patients. These findings call for continuous epidemiological surveillance and implementation of prevention strategies to reduce infections and dissemination of such a problematic pathogen inside the ICUs.
