Scaling-up molecular logic to meso-systems via self-assembly

Abstract Due to the small size and biocompatibility of molecules, molecular logic-based computation is a gateway to the informational basis of life processes. Logic-based computation operates widely with discrete molecules of up to nanometric sizes. The contribution of molecule-based bulk materials of milli/centimetric size to the field has begun in more recent years. However, artificial molecule-based meso-scale systems which intrinsically perform logic operations are very rare. Here, we show that self-assembled systems consisting of cyclophane octacarboxylates and a cationic surfactant can perform such functions, where a membrane itself behaves as a Reset-Set Flip-Flop which is integrated with 7 more logic elements. Now that molecular logic-based computation operates across a wide range of contiguous size-scales, the way opens for its general use in information processing aspects of biology and synthetic biology.

Abstract Carbon dioxide removal (CDR) is deemed necessary to attain the Paris Agreement’s climate objectives. While bioenergy with carbon capture and storage (BECCS) has generated substantial attention, sustainability concerns have led to increased examination of alternative strategies, including enhanced rock weathering (EW). We analyse the role of EW under cost-effective mitigation pathways, by including the CDR potential of basalt applications from silicate weathering (geochemical CDR) and enhanced ecosystem growth and carbon storage in response to phosphorus released by basalt (biotic CDR). Using an integrated carbon cycle, climate and energy system model, we show that the application of basalt to forests could triple the level of carbon sequestration induced by EW compared to an application restricted to croplands. EW also reduces the costs of achieving the Paris Agreement targets as well as the reliance on BECCS. Further understanding requires improved knowledge of weathering rates and basalt side-effects through field testing.

A functional screen uncovers circular RNAs regulating excitatory synaptogenesis in hippocampal neurons

Abstract Circular RNAs (circRNAs) are an expanding class of largely unexplored RNAs which are prominently enriched in the mammalian brain. Here, we systematically interrogate their role in excitatory synaptogenesis of rat hippocampal neurons using RNA interference. Thereby, we identify seven circRNAs as negative regulators of excitatory synapse formation, many of which contain high-affinity microRNA binding sites. Knockdown of one of these candidates, circRERE , promotes the formation of electrophysiologically silent synapses. Mechanistically, circRERE knockdown results in a preferential upregulation of synaptic mRNAs containing binding sites for miR-128-3p. Overexpression of circRERE stabilizes miR-128-3p and rescues exaggerated synapse formation upon circRERE knockdown in a miR-128-3p binding site-specific manner. Overall, our results uncover circRERE -mediated stabilization of miR-128-3p as a means to restrict the formation of silent excitatory synaptic co-clusters and more generally implicate circRNA-dependent microRNA regulation in the control of synapse development and function.
