Long term patterns and risk factors of loneliness in young adults from an 18-Year longitudinal study in Germany

Abstract Preventing loneliness is a significant goal due to its association with long-term negative effects on both mental and physical health. This German longitudinal study used three assessment points to examine predictors of emotional loneliness—defined as the absence of close, trusting relationships—in young adulthood (T3: age 22), based on factors assessed in early childhood (T1: age 4) and adolescence (T2: age 14). Utilizing data from 224 families, regression models were employed to identify child and parent risk factors—such as child and parent mental health problems, parent dysfunctional parenting, child adverse childhood experiences, child problematic internet use, and child experiences of school bullying—to predict child emotional loneliness. Emotional loneliness was measured through self-reports by adolescents/young adults and external reports provided by parents. Over 25% of adolescents reported emotional loneliness, a figure that increased to 50% in young adulthood during the pandemic. Experiencing loneliness in adolescence was associated with a significantly higher likelihood of feeling lonely again in emerging adulthood. Parents tend to underestimate their children’s feelings of loneliness. Lower levels of childhood externalizing mental health problems and higher levels of maternal dysfunctional parenting were associated with an increased likelihood of loneliness in adolescence. Only adolescent mental health problems predicted loneliness in emerging adulthood. Additionally, more adverse childhood experiences and higher levels of compulsive internet use during adolescence were linked to more adolescent mental health problems, identifying potential indirect targets for the prevention of loneliness. In contrast, paternal factors did not significantly contribute to the prediction of emotional loneliness in this sample. Effective strategies may include enhancing parenting practices and increasing awareness of child mental health problems, mitigating adverse childhood experiences, and reducing excessive internet use.

Abstract Targeting tumor proteostasis has emerged as a promising strategy in anticancer therapy, particularly through Hsp90 inhibition, which has shown clinical potential. However, the efficacy of Hsp90 inhibitors is limited by the activation of HSF1, a master regulator of the heat shock response (HSR), which mitigates proteotoxic stress by inducing protective chaperones. To address this limitation, we investigated the role of HSF1 SUMOylation in modulating its activity and its impact on Hsp90 inhibitor efficacy. We generated HSF1 mutants with lysine-to-arginine substitutions at five SUMOylation sites and studied their function in H1299 lung carcinoma cells with HSF1/HSF2 knockout, which lack a functional HSR. Unexpectedly, these mutants retained full transcriptional activity during the early phase of the heat shock response, mimicking the initial stress response of wild-type HSF1. SUMOylation inhibition using Subasumstat also led to altered nuclear stress bodies morphology but did not impair Hsp70 induction or enhance Hsp90 inhibitor cytotoxicity. Our findings reveal that SUMOylation is dispensable for HSF1 activation and transactivation capacity during the early phase of HSR. These results refine our understanding of HSF1 regulation and suggest that alternative strategies targeting HSF1 stability and degradation may enhance the therapeutic efficacy of proteostasis-targeting cancer therapies.

Abstract Airborne transmission of respiratory pathogens in indoor environments remains a significant global health challenge. While existing research broadly addresses ventilation effectiveness, there is a critical need to understand how specific diffuser placements influence early-phase aerosol dispersion immediately following a cough event. This study uses Computational Fluid Dynamics (CFD) with an Eulerian–Lagrangian approach and the Discrete Phase Model to analyze initial droplet transport, evaporation, and nuclei concentration under different air distribution configurations. The results demonstrate that conventional parallel exhaust configurations, though effective at reducing overall particle mass, can fail to control the lateral spread of infectious nuclei in the short term. In contrast, placing exhaust diffusers above the cough source reduces the lateral particle spread by approximately 40% compared to conventional layouts. Additionally, maintaining the WHO-recommended two-meter distance results in an 82–89% reduction in particle number concentration during the early dispersion phase. These findings underscore the importance of diffuser placement for controlling short-term particle dispersion immediately after a cough event in mechanically ventilated office environments. The study’s scope is limited to early-phase dispersion dynamics within a 10-second simulation period, and further research is needed to assess long-term aerosol suspension, removal mechanisms, and infection risk. Nonetheless, the results offer practical insights for HVAC design and support the integration of ventilation strategies with physical distancing measures to reduce near-field exposure risks.

Expression of pathogenesis-related proteins in potato brown rot plants confers resistance to filamentous pathogens under field trials

Abstract The present investigation aims to use innovative metagenomics technologies and denaturing gradient gel electrophoresis (DGGE) to compare the microbial communities of conductive and suppressive soils in connection to brown rot disease in the entire value of Egyptian potato imports. Besides evolution, the bioefficacy of two biocontrol agents, either alone or in consortium, on plant growth promotion and activation of defense responses in potato against the brown rot diseases. The soil status of collecting soil from seven different locations in different governorates was evaluated for tolerance to the artificial inoculation of the pathogen agent of R. solanacearum on potato plants. The DGGE uses 16 srRNA primers that were used to compare two extreme patterns for identifying the microbial population detected in environmental samples. Different bands were extracted from the DGGE gel and sequenced. The sequencing data results revealed that the biocontrol agent factor has a 100% gene bank similarity and belongs to the Pseudomonas species. The Pseudomonas nucleotide sequences isolates were deposited in the Gene Bank under Accession Nos. PP930812, PQ466864, and PQ470140. The findings showed that the soil from the governorates of Kerdasa exhibited a less tolerant treatment, with an estimated disease severity of 78.33%. On the other hand, ElBeheira Kom Hamada soil showed significant tolerance, with an estimated disease severity of 11.67. According to the data of gene expression analysis, both treated plants with the biocontrol agents showed a significant increase in PR-1, PR-2 and PR-Q gene expression, which accelerated the defense response and reduced brown rot disease. The evaluated potato genotype results proved to have the potential for brown rot disease resistance and higher yield production. The findings of this study recommended that biopriming with a microbial consortium enhance potato growth, productivity, and induction of defense responses against S. tuberosum through the induction of systemic resistance via expression of PR pathogenic-related protein network. The present investigation offers significant perspectives that warrant further exploration in subsequent studies to address the traceability requirements of the worldwide economics of disease control for potato plants.
