Infrared and visible single-pixel holographic microscope

Abstract

Holographic microscopy, which enables simultaneous observation of amplitude and phase information from label-free microscale samples, plays a crucial role in biomedical research and materials science. However, until now, holographic microscopy has been largely restricted to imaging in the visible region. Here, we present a holographic microscope single-pixel coherent detection to reconstruct amplitude and phase images in the visible and shortwave infrared simultaneously. We first validated the microscopy system's high-resolution capability of 2.46 μm and applied it to various biological samples. By employing milk as a surrogate scattering medium, we demonstrated the superior scattering robustness of the shortwave infrared. Concurrently, visible light showed enhanced efficacy in amplitude and phase reconstruction of transparent samples, thereby highlighting the complementarity of the visible and shortwave infrared. Moreover, under a 25% down-sampling strategy, the imaging frame rate reached 5.54 Hz at a resolution of 64 × 64 pixels.
