Research on the evolution and evaluation of Chinese sports industry policies based on intelligent analysis
In recent decades, with the support and traction of a number of key policy steps, the scale of China’s sports industry has achieved a new leap. The optimization of industrial structure has made new progress. From "nascent" to "strong", China’s sports industry grows in importance of the national economy. In the meantime, sport is a significant way to promote health. With the rapid growth of people’s requirements for sport and health, it is urgent to re-evaluate the past development path and formulate new directions so as to continuously improve and optimize the system. This study systematically sorts out China’s sports industry documents at different stages, and describes the focus of each stage and the overall evolution track. On this basis, text mining and quantitative evaluation being used to extract high-frequency words of sports industry documents, and a sports industry document evaluation system including 9 first-level indicators and 47 second-level indicators is established. In this study, text similarity analysis is used to realize intelligent PMC index analysis, which effectively improves the analysis efficiency and makes up for the deficiency of simple qualitative analysis. According to the study, China’s sports industry policies are scientific and effective. Combining with the development direction of industrial transformation, it provides ideas for the future adjustment and optimization of sports industry evolution path.

A postzygotic GNA13 variant upregulates the RHOA/ROCK pathway and alters melanocyte function in a mosaic skin hypopigmentation syndrome
Abstract The genetic bases of mosaic pigmentation disorders have increasingly been identified, but these conditions remain poorly characterised, and their pathophysiology is unclear. Here, we report in four unrelated patients that a recurrent postzygotic mutation in GNA13 is responsible for a recognizable syndrome with hypomelanosis of Ito associated with developmental anomalies. GNA13 encodes Gα13, a subunit of αβγ heterotrimeric G proteins coupled to specific transmembrane receptors known as G-protein coupled receptors. In-depth functional investigations revealed that this R200K mutation provides a gain of function to Gα13. Mechanistically, we show that this variant hyperactivates the RHOA/ROCK signalling pathway that consequently increases actin polymerisation and myosin light chains phosphorylation, and promotes melanocytes rounding. Our results also indicate that R200K Gα13 hyperactivates the YAP signalling pathway. All these changes appear to affect cell migration and adhesion but not the proliferation. Our results suggest that hypopigmentation can result from a defect in melanosome transfer to keratinocytes due to cell shape alterations. These findings highlight the interaction between heterotrimeric G proteins and the RHOA pathway, and their role in melanocyte function.
Carsharing operation optimization with the comprehensive consideration of economic and social benefits
Carsharing is regarded as a new mode of transportation that can meet the diversity of travel demands. This study develops a bi-level programming model to design operating strategies aiming to maximize economic and social benefits by determining the site location, number, size and carsharing pricing. In the upper model, a new mixed integer linear programming model is pro-posed to plan the site location, number, size, and pricing of carsharing jointly. Based on the stochastic user equilibrium principle, the lower model is a variational inequality model of travel mode and route choice, to understand the evolution of the transportation system structure after the introduction of shared cars. The results of the case study show the designed scheme can increase the economic benefits of carsharing enterprises and the social benefits of the government, and the site location, number, size, and pricing scheme impact benefits significantly. Furthermore, the setting of shared car sites at public transfer points can promote the interaction between shared cars and other modes of transportation. This study is helpful for carsharing enterprises to increase their economic benefits, enhance their capacity for sustainable growth, and improve the accessibility and flexibility of urban public transportation through sound operational strategies.

Selecting indicators for the measurement of low-value care using German claims data: A three-round modified Delphi panel
By reducing healthcare services that offer little benefit or potential harm to patients (low-value care), resources can be redirected towards more adequate treatments, improving healthcare efficiency and patient outcomes. This study aimed to systematically incorporate clinical expertise across medical disciplines through a Delphi process to establish indicators for measuring low-value care, ensuring their acceptance by medical societies, the broader medical community, and patients. We developed two versions (one with higher sensitivity and one with higher specificity) for almost each of the 42 indicators identified as potentially measurable in a previous systematic review. We conducted a three-round modified Delphi panel based on the RAND/UCLA appropriateness methodology, with 62 experts from 52 Scientific Medical Societies and professional organisations, and patient representatives. In round one, each indicator was rated for its ability to indicate low-value healthcare and its measurability in German claims data. This was followed by an online discussion in round two. The indicators were then modified based on expert feedback and re-assessed in round three. As a result, 24 indicators were deemed appropriate for measuring low-value care, covering areas such as pharmaceuticals, diagnostic tests, screening, and treatment. For example, one indicator identified patients with cancer who received chemotherapy in the last month of life. These indicators will help identify healthcare services that may require policy-level interventions to improve the quality of care. However, most low-value care indicators can only be measured in German claims data if documentation requirements for relevant information are expanded.

Prevalence of burnout syndrome in Brazilian anesthesiologists during the COVID-19 pandemic: A cross-sectional survey
Background Burnout syndrome, one of the consequences of chronic exposure to stressful, is more prevalent among physicians compared to the general population. Anesthesiology, alongside high-stress specialties such as emergency medicine and surgery, is particularly susceptible to this condition. During the COVID-19 pandemic, anesthesiologists were often on the front lines, potentially exacerbating burnout. This study aimed to assess the prevalence of burnout syndrome among Brazilian anesthesiologists during the pandemic. Methods A cross-sectional analytical observational study was conducted with all members of the Brazilian Society of Anesthesiology (SBA). Data were collected via sociodemographic questionnaires and the Maslach Burnout Inventory (MBI), disseminated by email. Results Burnout syndrome was identified in 19.6% (n = 213) of respondents, while 56.5% (n = 613) were at high risk for developing burnout. Having considered quitting the specialty was the variable most strongly associated with the prevalence of burnout syndrome and the high risk of burnout. As a protective factor, dedicating more time to leisure (over 5 hours per week) was related to a lower occurrence of burnout syndrome and its risk. Conclusion Burnout syndrome is highly prevalent among Brazilian anesthesiologists and residents. Target strategies to mitigate burnout should be implemented by healthcare institutions, professional organizations, and government bodies.

Effect of using home-based dynamic intermittent pneumatic compression therapy during periods of physical activity on functional and vascular health outcomes in chronic stroke: A randomized controlled clinical trial
Background Intermittent pneumatic compression (IPC) therapy may benefit stroke patients by eliciting more intensive training sessions that may result in better health, mobility and ultimately quality of life. The purpose of this randomized controlled trial was to assess the effect of using a home-based IPC device on functional outcomes and vascular health in individuals with chronic stroke. Methods Thirty-one stroke survivors (64.3 ± 14.3y; 4.3 ± 2.7y since stroke) completed pre- and post-intervention assessments of functional capacity (six-minute walk test [6MWT], timed-up-and-go, 10m walk test), vascular health (pulse wave analysis, carotid-femoral pulse wave velocity), and physical activity. Following the pre-assessment, individuals were randomly assigned to either a daily, 12-week, home-based IPC group, or to a usual care control (CON) group. Outcomes were assessed using analysis of covariance (ANCOVA), controlling for age and any baseline differences. Results Following ANCOVA, a significant increase in 6MWT walking distance was observed post-assessment for the IPC (Mean ± SD [95%CI]; 188 ± 19 m [177–199m]) but not the CON group (167 ± 19 m [157–178m]) (p < 0.05). A significant reduction in peripheral systolic blood pressure was reported at the post-assessment for the IPC group (136.2 ± 8.0 mmHg [131.9–140.4 mmHg]) but not for CON (142.2 ± 8.0 mmHg [138.1–144.6 mmHg]) (p < 0.05). Similar findings were observed for central systolic blood pressure. Physical activity levels significantly increased at the post-assessment for IPC (1857 ± 879 MET·min−1·week−1 [1390–2325 MET·min−1·week−1]) but not for the CON group (1161 ± 879 MET·min−1·week−1 [677–1645 MET·min−1·week−1]), while for time spent sitting, a significantly greater reduction was observed at the post-assessment for the IPC group (396 ± 86 mins [350–442 mins]) compared to CON (486 ± 86 mins [439–534 mins]) (both p < 0.05). Conclusions The observed improvements in functional mobility, cardiovascular health, increased physical activity and reduced sedentary time demonstrates important clinical implications of ‘home-based’ IPC therapy as a clinical training aid for stroke rehabilitation. Home-based IPC therapy could serve as an adjunct to conventional rehabilitation, however, further research is needed to determine whether IPC therapy can sustain or improve function over time for individuals in the chronic stage of recovery.

The canonical RPA complex interacts with Est3 to regulate yeast telomerase activity
In most eukaryotic organisms, cells that rely on continuous cell division employ the enzyme telomerase which replenishes chromosome termini through the addition of telomeric repeats. In budding yeast, the telomerase holoenzyme is composed of a catalytic core associated with two regulatory subunits, Est1 and Est3. The Est1 protein binds a telomere-specific RPA-like complex to recruit telomerase to chromosome ends. However, the regulatory function of the Est3 subunit has remained elusive. We report here that an interaction between Est3 and the canonical RPA complex is required for in vivo telomerase function, as revealed by mutations in RPA2 that confer an Est (Ever shorter telomeres) phenotype, characteristic of a defect in the telomerase pathway. Binding between RPA and telomerase, which is supported by compensatory charge-swap mutations in EST3 and RPA2 , utilizes a surface on Est3 that is structurally analogous to an interface on the human TPP1 protein that is required for telomerase processivity. Mutations in a subset of conserved DNA contact residues in RPA also result in short telomeres and senescence, which we show is due to a requirement for DNA binding after RPA interacts with telomerase. We propose that once RPA forms a complex with telomerase, RPA utilizes a subset of DNA-binding domains to stabilize the interaction between the telomerase active site and telomeric substrates, thereby facilitating enzyme processivity. These results, combined with prior observations, show that yeast telomerase interacts with two different high-affinity ssDNA-binding complexes, indicating that management of single-stranded DNA is integral to effective telomerase function.

Targeting beta-lactamase activity with Oxacyclohexadecan-2-one in carbapenem-resistant uropathogenic E. coli: A molecular simulation approach
Urinary tract infections caused by uropathogenic Escherichia coli (E. coli) are a global health concern, with rising rates and antibiotic resistance demanding novel treatments. Therefore, in this study, we explored the potential of Oxacyclohexadecan-2-one obtained from Moringa oleifera (M. oleifera) seed, as antibacterial agent against three majorly prevalent carbapenemase-producing E. coli proteins, blaNDM-1 (New Delhi metallo-betalactamase-1), blaNDM-5 (New Delhi metallo-betalactamase-5) and blaOXA-48 (Oxacillinase-48) from the strains Ecw3, EC-114 and T20 respectively. The ethanolic extract of M. oleifera seed was subjected to GC-MS, identifying 135 compounds. PyRx virtual screening, identified the top 10 ligands for each protein following the Rule of 5 and ProTox classes V and VI, with Oxacyclohexadecan-2-one (PubChem ID: 235414) showing best binding affinity across all 3 proteins with an optimized dose (LD50) of 5000mg/kg. Hence, molecular docking was carried out for ligand 235414 along with Imipenem, belonging to the same class V toxicity class with an optimized dose (LD50) of 5000mg/kg. Imipenem is a commonly used FDA drug to treat UTIs, which served as the control in the study. Oxacyclohexadecan-2-one showed higher binding affinity for the beta-lactamase proteins with a docking score of -6.45 kcal/mol, -6.05 kcal/mol and -7.34 kcal/mol compared to -3.41 kcal/mol, -3.99 kcal/mol and -6.36 kcal/mol of Imipenem for NDM-1, NDM-5 and OXA-48 respectively. Dynamic Simulation was performed for 100 ns for Oxacyclohexadecan-2-one and Imipenem bound protein complexes to determine the stability, fluctuations, compactness, bond interaction, solvent accessibility area, free energy landscape and the binding free energy. The results of molecular docking and dynamics were promising for the Oxacyclohexadecan-2-one, suggesting its potent inhibitory effect against the beta-lactamase producing proteins.

A humanized NOVA1 splicing factor alters mouse vocal communications
Abstract NOVA1, a neuronal RNA-binding protein expressed in the central nervous system, is essential for survival in mice and normal development in humans. A single amino acid change (I197V) in NOVA1’s second RNA binding domain is unique to modern humans. To study its physiological effects, we generated mice carrying the human-specific I197V variant (Nova1 hu/hu ) and analyzed the molecular and behavioral consequences. While the I197V substitution had minimal impact on NOVA1’s RNA binding capacity, it led to specific effects on alternative splicing, and CLIP revealed multiple binding peaks in mouse brain transcripts involved in vocalization. These molecular findings were associated with behavioral differences in vocalization patterns in Nova1 hu/hu mice as pups and adults. Our findings suggest that this human-specific NOVA1 substitution may have been part of an ancient evolutionary selective sweep in a common ancestral population of Homo sapiens, possibly contributing to the development of spoken language through differential RNA regulation during brain development.
Multi-agent deep reinforcement learning-based robotic arm assembly research
Due to the complexity and variability of application scenarios and the increasing demands for assembly, single-agent algorithms often face challenges in convergence and exhibit poor performance in robotic arm assembly processes. To address these issues, this paper proposes a method that employs a multi-agent reinforcement learning algorithm for the shaft-hole assembly of robotic arms, with a specific focus on square shaft-hole assemblies. First, we analyze the stages of hole-seeking, alignment, and insertion in the shaft-hole assembly process, based on a comprehensive study of the interactions between shafts and holes. Next, a reward function is designed by integrating the decoupled multi-agent deterministic deep deterministic policy gradient (DMDDPG) algorithm. Finally, a simulation environment is created in Gazebo, using circular and square shaft-holes as experimental subjects to model the robotic arm’s shaft-hole assembly. The simulation results indicate that the proposed algorithm, which models the first three joints and the last three joints of the robotic arm as multi-agents, demonstrates not only enhanced adaptability but also faster and more stable convergence.
