An insight into the salivary gland content of the human body louse, Pediculus humanus

Abstract Human body lice, Pediculus humanus humanus, are blood-feeding parasites that live in clothing and feed several times per day. Saliva injected during louse feeding induces pruritis and local inflammation in the skin. If untreated, chronic Pediculosis can cause systemic negative health effects. Despite the medical importance of body lice and their longstanding association with humans, characterization of their saliva has been limited. To address this, we extracted RNA and protein from two of the body louse’s morphologically distinct sets of salivary glands (Bean-shaped and U-shaped) and generated transcript and protein profiles for each. Additionally, we performed fluorescent staining and confocal microscopy on each gland type to enhance descriptions of their structure and gross cellular architecture. Analysis of body louse salivary gene products and proteins revealed that the overwhelming majority were not closely related to biomolecules of known function, highlighting the organism’s unique and understudied saliva composition. Despite the contrasting morphology of the two gland types, there was a high degree of overlap in the salivary products produced. This finding suggests strong Darwinian selection pressure to maintain both salivary gland types, given that it would be simpler to have a single morphologically identical set of glands. Here we present the first next-generation sequencing and proteomic characterization of the human body louse sialome, discuss the potential physiological importance of louse salivary proteins, and consider possible explanations for why lice have a complex salivary gland system despite inordinate redundancy in the protein repertoire of the Bean- and U-shaped salivary glands.

Molecular Stiffening by Macrocycle Clustering

Abstract Allosteric stiffening of a portion of a protein surface is a strategy used in nature to regulate protein oligomerization and provide crucial functions for cells. However, a similar strategy to selectively control part of a compound dynamics remains elusive. Here we show that cucurbit[ n ]uril (CB[ n ]) macrocycles can bind almost all portions of a tetratopic guest molecule, stiffening the different parts of the guest to different extents. “Host–guest” interactions were found to be instrumental in selectively “freezing” guest molecular motions. The combination of 1 H‐NMR (1D, 2D), DOSY, VT‐NMR, isothermal titration calorimetry (ITC), mass spectrometry and molecular modelling enabled to highlight the crucial role of cucurbit[8]uril (CB[8]) binding in the selective hardening of relevant portions of the guest molecule. Beyond implications for bioinspired systems mimicking control of a system dynamic to create a new function, this approach has relevance for improving room temperature phosphorescence, and could also be used to allosterically control organocatalysis in water.

Abstract Aquaporins are natural proteins that rapidly transport water across cell membranes, maintaining homeostasis, whilst strictly excluding salt. This has inspired their use in water purification and desalination, a critical emerging need. However, stability, scalability, and cost have prevented their widespread adoption in water purification membrane technologies. As such, attention has turned to the use of artificial water channels, with pore‐functionalized polymers and macrocycles providing a powerful alternative. Whilst impressive rates of transport have been achieved, the combination of a scalable, high‐yielding synthesis and efficient transport has not yet been reported. Herein, we report such a system, with densely functionalized channel interiors, synthesized by high‐yielding living polymerization with low polydispersities, showing high salt exclusion and excellent water transport rates. Our aramid foldamers create artificial water channels with hydrophobic interiors and single‐channel water permeability rates of up to 10 8 water molecules per second per channel, approaching the range of natural aquaporins (c. 10 9 ). We show that water transport rates closely correspond to the helical length, with the polymer that most closely matches bilayer thickness showing optimal efficacy, as supported by molecular dynamics (MD) simulations. Our work provides a basis for the scalable synthesis of next‐generation artificial water channels.

Improving Internal Exciton Confinement for Efficient CdZnSeS‐Based Blue Quantum Dot Light‐Emitting Diodes

Abstract Quantum‐dot light‐emitting diodes (QLEDs) are thought to be the base for next‐generation display technology. However, the performance of blue‐emitting QLEDs still falls behind those of green and red ones, which can be attributed to the energy loss from Auger recombination and the strong coupling of excitons with surface states. Blue quantum dots (QDs) with giant CdZnSeS alloy cores are expected to improve the internal confinement of excitons due to the nonmonotonical energy landscape of their conduction band, while they haven't shown high performance in QLEDs due to insufficient optimization of their shell structures. In this work, giant CdZnSeS alloy cores were synthesized by diffusing Zn atoms into CdSeS cores, so that the core/shell lattice stress was released due to the optimized gradient compositions. As a result, exciton transfer and Auger recombination are both suppressed, leading to a breakthrough external quantum efficiency (EQE) of 24% in blue QLEDs with giant CdZnSeS alloy cores. Compared to the more extensively studied blue quantum dots with CdSeZn alloy cores, blue QLEDs with giant CdZnSeS alloy cores also benefit from the suppressed Fermi level and nonmonotonical energy landscape of the conduction band minimum (CBM), which are crucial for confining the wavefunctions of the excitons. The improved exciton confinement explained the superior performances of giant CdZnSeS alloy cores over CdSeZn cores in blue QLEDs.

Cobaloxime‐Based Metalloradical Catalysis: A Versatile Strategy for the Synthesis of Cyclopropenes and Oxazoles

Abstract Metalloradical catalysis (MRC), predominantly exemplified by metalloporphyrin complexes, has emerged as a promising strategy for regulating radical reactions and broadening their synthetic applications. Here, an efficient catalytic method has been devised for the radical [2 + 1] cyclopropenation of alkynes and radical [3 + 2] oxazolation of nitriles using α‐aryldiazoacetates via MRC. Cobaloxime proves effective in facilitating the metalloradical [2 + 1] cyclopropenation of alkynes and radical [3 + 2] oxazolation cycloaddition reaction under mild conditions. The reactions demonstrate broad substrate tolerance, accommodating gram‐scale transformations and the synthesis of pharmaceutical compounds. Our findings highlight the significant potential of cobaloxime‐catalyzed cycloaddition as a valuable tool in organic synthesis and broaden the scope of metalloradical systems in catalysis.

Constructing Mechanical–Chemical Stability via Multiphase Riveting and Interface Optimization Toward Layer‐structured Oxide Cathode Material

Abstract Manganese‐based layer‐structured oxide materials are considered as one of the most competitive cathode materials for sodium‐ion batteries due to their low cost and efficient sodium intercalation chemistry. Their electrochemical performance, however, is hindered by mechanical and chemical failures stemming from weak interlayer interactions, the Jahn–Teller effect of Mn 3+ and unstable surfaces. To address these issues, a quenching method was employed to fabricate a robust multiphase structure with a fluorine and dislocation‐rich surface. Through the accumulation of dislocations and the interlocking of multiphase structures, the mechanical stability of the material during (de)sodiation processes is enhanced, while the surface fluorine anchoring further strengthens the chemical stability. Even after 200 cycles at 0.5 C and 1 C within the voltage range of 1.5–4.5 V, the designed composite material P2/P3/O3‐Na 0.89 Ni 0.3 Mn 0.55 Cu 0.1 Ti 0.05 O 1.94 F 0.06 exhibits impressive capacity retention rates of 87.17% and 90.4%, respectively. This work exemplifies the important role of simultaneous design of mechano‐chemically coupled materials for the development of high performances cathode materials.
