Online Coupling High‐Temperature Electrolysis with Carbonylation Reactions: A Powerful Method for Continuous Carbon Dioxide Utilization

Abstract Despite being widely available, the broad utilization of CO 2 for chemical production remains in its infancy. The difficulty of using CO 2 as an important resource for the chemical industry lies in the high stability of the molecule and its associated inertia. In this work, we demonstrate how to overcome these limitations by utilizing a solid oxide electrolysis cell (SOEC) with an optimized cathode to produce dry CO through the high‐temperature electrolysis of CO 2 . We further integrate this process with a synthesis setup in a continuous‐flow coil reactor to boost reactivity in various carbonylation processes, including in amino‐, alkoxy‐, and phenoxycarbonylation, carbonylative Sonogashira couplings, or the synthesis of redox‐active esters. Ultimately, our approach offers a platform strategy for the rapid, scalable, and continuous production of carbonyl compounds directly from CO 2 while eliminating the requirement for storing large CO quantities.

Abstract Thin film is the form of material that most closely resembles the silicon-based integrated circuits (IC) and therefore has attracted tremendous attention over the past decades due to its potential applications in integrating functional devices on IC chips. The structural characterization of thin films, especially epitaxial film with complex structure has been a long-term challenge until the emergence of synchrotron three-dimensional diffraction technique (3D-RSM). 3D-RSM is a technique that can effectively collect various structural information of epitaxial films, such as crystal lattice, strain, domain variants, and oxygen octahedral rotation. Now, interpreting the massive experimental data of 3D-RSM becomes the biggest obstacle that is confronted by the researchers. In this work, we proposed a strategy that utilizes simulated 3D-RSM diffraction patterns as aid of data analysis. With this approach, the one-to-one correspondence between diffraction spots and domain variants, as well as the quantitative lattice constants and crystal system have been identified in sequence for two typical cases, either epitaxial PbTiO3/SmScO3(001) film or (CoCrFeMnNi)3O4 alloy film epitaxially grown on LaAlO3(001) substrate. Further, systematic simulations of 3D-RSM patterns for epitaxial films belonging to every of the seven crystalline symmetries were performed and exhibited, assuming the films are grown on a (001)-oriented cubic substrate. This work sheds light on more effective data analysis of 3D-RSM, i.e., more effective structural characterization of complex epitaxial films.

Construction of Multistep Charge Transfer Pathways in Bi <sup>0</sup> @Bi <sup>3+</sup> ‐KNbO <sub>3</sub> for Significantly Accelerated Photoconversion of Waste Plastics

Abstract Photoconversion of waste plastics into valuable CO and CH 3 COOH represents a ground‐breaking strategy for addressing plastic pollution issues. However, this process currently encounters significant challenges, primarily due to the limitation of catalyst activity and the difficulty in breaking C─C bonds. Herein, we present a novel approach that integrates multistep charge transfer pathways with photothermal‐driven reactions to improve photoconversion efficiency. By incorporating Bi 0 /Bi 3+ metal as an electron transport mediator for multistep charge transfer, we markedly enhanced the separation and transport of photoelectrons, thereby accelerating the generation of active species. Meanwhile, the heat generated by the localized surface plasmon resonance effect of Bi 0 drove the reactions related to the photoconversion of polypropylene. Subsequently, the photoconversion rates of PP into CO by Bi 0 @Bi 3+ ‐KNbO 3 reached 209.41 µmol g cat −1  h −1 , which is 27.55 times higher than that achieved with KNbO 3 . Furthermore, the dual Bi–Nb sites effectively stabilize the key intermediate *COOH, thereby promoting the production of CH 3 COOH at a rate of 213.00 µmol g cat −1  h −1 . This strategy of boosting photoconversion activity of PP into CO and CH 3 COOH offers an effective green solution to the serious issue of plastic pollution.

Enantio‐ and Diastereoselective Synthesis and Spiral‐Stair‐Like Single Helix Assembly of Figure‐Eight Cyclophenylenes

Abstract Helix assemblies of chiral molecules can transfer microscopic unimolecular chirality to macroscopic supramolecular chirality, enhancing various chiral properties. In addition to the commonly observed spiral‐column‐like helix assembly, a small number of spiral‐stair‐like helix assemblies have also been reported in aromatic nanocarbons with multiple chirality‐related irregularities. However, they require separation of diastereomers and/or enantiomers or do not have stable chirality. Here, we report the enantio‐ and diastereoselective synthesis of figure‐eight [10]cyclophenylenes with stable helical chirality by the rhodium‐catalyzed four consecutive intramolecular [2 + 2 + 2] cycloadditions of dodecaynes with two flexible biphenyl units. The chiral figure‐eight [10]cyclophenylene with ethyl and methyl side chains exhibits the spiral‐stair‐like single helix assembly in the crystal due to CH–π and CH–O interactions and good CPL properties in solution. Experimental verification of the enantio‐ and diastereodetermining steps of four consecutive [2 + 2 + 2] cycloadditions is also reported.

High-fidelity in silico generation and augmentation of TCR repertoire data using generative adversarial networks

Abstract Engineered T-cell receptor (eTCR) systems rely on accurately generated T-cell receptor (TCR) sequences to enhance immunotherapy predictability and efficacy. The most variable and crucial part of the TCR receptor is the CDR3 sequence region. Current methods for generating CDR3 sequences, including motif-based and Markov models, struggle to generate reliable, diverse, and novel TCR sequences. In this study, we present the first application of Generative Adversarial Networks (GANs) for producing biologically reliable CDR3 sequences, using Long Short-Term Memory (LSTM) networks and LeakyReLU-based GANs. Our results show that LSTM models generate more diverse sequences with higher accuracy, lower discriminator loss, and higher AUC compared to LeakyReLU. However, LeakyReLU provides greater stability with a lower generator loss, achieving a total Pearson correlation score of over 0.9. Both models demonstrate the ability to produce highly realistic TCR sequences, as validated by t-SNE clustering, frequency distribution analysis, TCRd3 BLAST analysis, and in silico docking. These findings highlight the potential of GANs as a powerful tool for generating synthetic yet biologically relevant TCR sequences, a crucial step toward improving eTCR-based therapies. Further refinement of amino acid frequency distributions and clinical validation will enhance their applicability for therapeutic purposes.

Rapid Access to Conjugated <i>Z</i> , <i>Z</i> , <i>Z</i> ‐Trienes

Abstract Intercepting reactions have recently emerged as an innovative approach in organic synthesis, allowing chemists to harness reactive intermediates to access structures and functionalities that conventional methods cannot easily achieve. By deliberately manipulating reaction pathways, this strategy provides a unique avenue to explore unusual reactivities and complex molecular architectures. In this work, we have developed a new protocol for the nucleophile‐intercepted Beckmann fragmentation reaction (NuBFr). Employing tropone oxime tosylate – a simple and readily accessible precursor – and a diverse set of nucleophiles generated in situ from alcohols, phenols, thiols and alkynes with strong bases, we successfully synthesized a library of novel conjugated Z , Z , Z ‐trienecarbonitrile derivatives. This method facilitates rapid access to the conjugated Z , Z , Z ‐triene motif, a structural feature rarely encountered in synthetic chemistry. Our computational studies indicate that the NuBFr reaction likely proceeds through the formation of a bicylic azirine intermediate. The resulting Z , Z , Z ‐trienecarbonitriles could undergo an unprecedented thermally induced 8π/6π/4π electrocyclization cascade sequence to produce trisubstituted olefins. This sequence underscores the fundamental value of these motifs in pushing the boundaries of unusual reactivities in organic synthesis.

Cross‐Linking Profiling of Molecular Glue Degrader‐Induced E3 Ligase Interactome to Expand Target Space

Abstract Molecular glue (MG) degraders, small molecules with significant therapeutic potential for targeting undruggable proteins, are emerging as new modality in drug discovery. Profiling the E3 ligase interactome induced by MG degraders provides insights into their mechanism of action and identifies clinically relevant neosubstrates for degradation, thereby offering new therapeutic opportunities. However, established methods face significant challenges in comprehensive and accurate profiling of MG degrader‐induced E3 ligase interactome. Herein, we introduce the concept of globally cross‐linking profiling of the MG degrader‐induced E3 ligase interactome in living cells, achieved by integrating genetic code expansion technology with mass spectrometry‐based proteomics. Our approach presents an efficient and robust strategy for identifying neosubstrates recruited to cereblon E3 ligase by the known degraders CC‐885 and DKY709, offering valuable insights for clinical evaluation and significantly expanding their target space. Moreover, we developed two novel MG degraders with potent antiproliferative effects on cancer cells, and application of our method identified neosubstrates, revealing a previously unrecognized target landscape and advancing our understanding of E3 ligase–neosubstrate interactions. Overall, our study provides a powerful tool for neosubstrate identification and expanding target space of E3 ligase, opening new opportunities for developing next‐generation MG degraders to address the clinical challenge of undruggable targets.
