Protocatechuic acid inhibits aflatoxin production and modulates gene expression in Aspergillus flavus
Abstract Aflatoxins, particularly Aflatoxin B1 (AFB1), produced by Aspergillus flavus and other species, pose significant health risks due to their carcinogenic properties. This study investigates the inhibitory effects of Protocatechuic Acid (PCA) on mycotoxigenic fungi and AFB1 production. PCA demonstrated significant dose-dependent antifungal activity against various Aspergillus species, with A. flavus showing inhibition zones ranging from 5.3 mm to 16.7 mm at concentrations of 50 µg/ml to 250 µg/ml, while A. ochraceus exhibited the highest sensitivity, with zones up to 23.6 mm. Additionally, PCA effectively reduced AFB1 production in liquid media, achieving up to 80.21% inhibition at 250 µg/ml, and decreased the mycelial weight of A. flavus by 60.8%. Gene expression analysis revealed that PCA significantly downregulated the expression of the AFB1 biosynthetic genes nor-1 (95% reduction) and omt-A (74% reduction), suggesting that PCA disrupts multiple stages of aflatoxin synthesis. Furthermore, PCA demonstrated efficacy in controlling AFB1 contamination in postharvest corn grains, with inhibition percentages of 44.8%, 55.7%, and 64.6% at 150, 200, and 250 µg/ml, respectively. These findings indicate PCA’s potential as a natural antifungal agent, offering promising applications in food safety and postharvest management.

Abstract This paper presents a comparative analysis of the impact of individual elements of the W14 fastening system on clamping force. Clamping force in the context of railway fastening systems refers to the force that holds the rail firmly against the sleeper to ensure track stability and maintain proper rail alignment under dynamic loads. The results of laboratory tests carried out in accordance with the applicable European standards of the PN-EN 13,481 and PN-EN 13,146 series are described synthetically. Actual clamping force achieved is highly dependent on the height of the rail pad, as the applied torque can result in different clamping forces depending on this factor. It is important to note that the critical parameter for the fastening system’s performance is not the applied torque itself, but rather the resulting clamping force. This force is directly influenced by the rail pad height, as well. as other factors such as the static stiffness of the pad. Therefore, achieving the appropriate clamping force requires careful consideration of these factors, as they significantly impact the system’s stability and overall efficiency.

Childhood predictors of religious reading: a cross-national analysis in the Global Flourishing Study
Abstract While the family environment, including parental religiosity, shapes children’s religious behaviors, evidence concerning factors influencing religious reading and listening (RR/L) in adulthood remains scarce. This study examines 13 early-life conditions within familial and economic contexts as potential predictors of adult RR/L. Nationally representative samples from 22 countries (N = 202,898) included in the first wave of the Global Flourishing Study were used. Multiple imputation, multivariate regression analyses, and random-effects meta-analyses were employed to evaluate associations between the childhood predictors and adult RR/L. Meta-analytical results show that early-life predictors, including religious service attendance, a positive relationship with one’s father, excellent self-assessed health, experience of abuse, and growing up as an outsider, as well as belonging to older cohorts and being female are associated with higher likelihood of adult RR/L, with religious service attendance being the strongest predictor. There was little evidence of associations with childhood financial status, immigration status, and mother-child relationship. Country-specific analyses revealed that race/ethnicity and religious affiliation also influence the RR/L likelihood, with Muslims and minority group members showing a higher probability compared to other groups. Given the rare examination of early-life determinants of RR/L, especially in cross-cultural contexts, this study provides valuable insights into the global distribution of early-life predictors of RR/L.

Ptychoscopy: a user friendly experimental design tool for ptychography
Abstract Electron ptychography is a rapidly growing diffractive imaging technique providing superior image contrast, high resolution, efficient dose usage and versatile imaging conditions. Originally coming from the physical sciences, it has also become a method of interest in the life sciences. Regardless of the scientific field, the successful ptychographic reconstruction relies on various combinations of adjustable experimental parameters, along with the choice of the reconstruction algorithm. In this work we present ptychoScopy, a Python-based tool developed to simplify the selection of these parameters and aid successful experimental design. Using a $$\hbox {SmB}_{6}$$ sample, we show the influence of key parameters such as probe convergence angle, defocus and electron dose on the reconstruction quality using direct and iterative reconstruction algorithms. We investigate the influence of real and reciprocal space sampling and their influence on the speed and quality of ptychographic reconstructions. PtychoScopy simplifies experimental design and guides the researchers across diverse scientific fields in setting up successful ptychographic experiments.
