Estuaries of the Tinto, Odiel and Piedras rivers as source of new species of Pseudomonas with biofertilizer potential under stress conditions

Abstract The estuaries of the Tinto, Odiel, and Piedras rivers have been extensively studied by several groups due to ecological aspects such as heavy metal contamination, nutrient deficiency and degradation influenced by climate change. Aimed to recover these ecosystems, a large collection of plant growth-promoting bacteria (PGPB), several of them previously identified as new species, was isolated from the rhizosphere and tissues of native plants. In this study, we analyse the strains N4, N8 T , L1 T , and SDT3 T as potential new species. A phylogenomic analysis revealed that four strains belong to the genus Pseudomonas , forming distinct clades with full support. Digital DNA-DNA hybridization and average nucleotide identity values were below the species delineation thresholds (70% and 95–96%), confirming the novelty of strains N8 T , L1 T , and SDT3 T , while N4 and N8 T were identified as the same species. As a result, we propose the names Pseudomonas medicaginis sp. nov., Pseudomonas onubensis sp. nov., and Pseudomonas spartinae sp. nov., with P. medicaginis comprising two strains. These strains, previously identified as PGPB, harboured genes related to PGP traits and were further characterized phenotypically and biochemically. To evaluate the agroecological potential of N8 T strain, it was tested in co-inoculation with the corresponding rhizobia on lentil, pea, alfalfa, and bean under greenhouse conditions. In all legumes, N8 T enhanced shoot and root biomass, increased nodule numbers, and improved nitrogen content compared with single rhizobia inoculation. In summary, these estuarine ecosystems are not only ecologically significant for their microbial diversity but also serve as reservoirs of beneficial bacteria with promising agroecological applications.

Cognitive alterations related to driving performance in Parkinson’s disease detected by a driving simulator

Abstract Driving ability in individuals with Parkinson’s disease (PD) can be compromised early in the course of the illness, even before clear cognitive deficits emerge on standard neuropsychological tests. This study investigated subtle driving impairments in a group of non-demented people with PD using a high-fidelity driving simulator. Seven PD participants and seven healthy controls, matched for age and sex, completed cognitive assessments, reaction time tasks, and five simulated driving scenarios that measured lane keeping, steering control, and reaction to events. While most cognitive scores were comparable between groups, PD participants exhibited slower response times in basic tasks and showed reduced lane control, particularly during left turns. These difficulties were associated with disease severity and medication dosage. The simulator proved more sensitive than conventional tests in detecting early impairments related to attention and visuospatial processing. These findings suggest that driving simulators may play a key role in improving the assessment of driving competence in PD, providing insight into real-world challenges faced by this population.

Burnout, depression, and medication errors among family physicians

Abstract Physician burnout has been recognized as a global healthcare threat. Burnout adversely affects doctors’ well-being, patient care costs, and quality. The impact of burnout on medication error is debatable since most literature relied on self-reporting, whereas studies utilizing objective measures reported contrary results. Our study investigates physicians’ burnout prevalence and association with detected prescribing errors in ambulatory settings. A cross-sectional study was conducted in 25 primary healthcare centers in Alexandria, Egypt. The Professional Fulfillment Index and Patient Health Questionnaire-9 were used to screen for burnout and depression among physicians. Clinical pharmacists independently reviewed 50 prescriptions per physician to identify decision and process prescribing errors. The prescribing errors were assessed by reviewing drug suitability for the patient’s condition according to best practice guidelines. Risk variables for prescribing errors were evaluated using multivariable negative binomial analysis. Researchers collected prescriptions from 118 doctors out of 184 who completed the survey. Almost half of the responders suffered from burnout and depression symptoms (42.39% and 42.93%), while most of them reported low personal professional fulfillment (89.13%). Of 14,121 medications evaluated, 6543 prescribing errors were detected (46.34%), with only 0.11% of them potentially serious. The most prevalent prescribing error types were wrong dosage regimens, incomplete patient information, and missing drug instructions. Analgesics, systemic antihistaminics, and antibacterials were the most misprescribed ATC drug categories. Prescribing errors had no significant association with burnout, professional fulfillment, depression, or thoughts of quitting among doctors. Males, consultants, and rural work settings physicians were more prone to commit ordering errors. Prescribing errors were prevalent in primary care settings, but they were not significantly linked to physicians’ burnout, professional fulfillment, or depression. The burden of burnout and depression among physicians, as well as the complexity of evaluating their impact on actual patient safety, highlights the need for further research and tailored interventions.

Allelopathic effects of tree peony extracts on bok choy, spinach and tatsoi

Abstract Although tree peony ( Paeonia ostii ) is widely cultivated in China, it has not been fully utilized as an arable resource. To investigate the suitability of tree peony in intercropping systems, we examined the allelopathic effects of aqueous extracts from the leaves and roots of P. ostii ‘Fengdan’ on the seed germination, growth, and physiological responses of tatsoi, spinach, and bok choy. Among these vegetables, tatsoi exhibited the best seed germination and seedling growth. Lower concentrations of aqueous extracts of ‘Fengdan’ roots (5–10 g L −1 for tatsoi and 5 g L −1 for spinach and bok choy) promoted seed germination, biomass, chlorophyll content, root activity, photosynthetic rate, intercellular CO 2 concentration, stomatal conductance, and antioxidant enzyme activity. However, high concentrations of the leaf extract (40 g L −1 for tatsoi and 20–40 g L −1 for spinach and bok choy) significantly increased levels of malondialdehyde and reduced seed germination, dry weight, chlorophyll content, root activity, and antioxidant enzyme activity. These deleterious effects increased with higher extract concentrations. The allelopathic effect of the leaf extract was greater than that of the root extract. The order of the allelopathic effect of tree peonies on the three vegetables was bok choy, spinach, and tatsoi. Therefore, tatsoi is the most suitable vegetable for interplanting with tree peonies.

Identification of potential biomarkers associated with mitochondrial oxidative stress in idiopathic pulmonary arterial hypertension via bioinformatic and experimental analysis

Abstract Idiopathic pulmonary arterial hypertension (IPAH) is a progressive and devastating disorder. Mitochondrial oxidative stress has been found to be involved in PAH development. This study aims to explore potential mitochondrial oxidative stress-related biomarkers in IPAH via integrated bioinformatics, validate these genes in MCT-induced rat PAH model and examine the functional roles of COX6B1 and HMGCL in mitochondrial oxidative stress in PDGF-induced pulmonary arterial smooth muscle cells (PASMCs). GSE15197 was downloaded from GEO database. Genes associated with mitochondria and oxidative stress were obtained from Molecular Signatures Database and MitoCarta3.0 databases. Weighted gene co-expression network analysis (WGCNA) was used. GO and KEGG enrichment analysis were utilized. Furthermore, machine learning algorithms (LASSO, SVM-RFE and RF) were constructed to screen hub genes. A nomogram model based on the hub genes were performed for evaluation the diagnostic value, and the expression of hub genes were validated by real-time quantitative PCR and Western blot in MCT-induced rat PAH model. Finally, COX6B1 and HMGCL specific siRNAs were transfected into PDGF-induced PASMCs, mitochondrial ROS (mtROS) level and MnSOD activity were examined. A total of 18 genes related to mitochondrial oxidative stress were identified in IPAH. Two hub genes (COX6B1 and HMGCL) were finally determined as potential biomarkers. Based on the 2 genes, a nomogram model was constructed. This model exhibited remarkable diagnostic efficiency values for IPAH. The expressions of COX6B1 and HMGCL were found to be upregulated in MCT-induced PAH model, in agreement with the findings of bioinformatics analysis. While, transfection with either COX6B1 siRNA or HMGCL siRNA suppressed the abnormal accumulation of mtROS and restored MnSOD activity in PDGF-induced PASMCs. Our results indicated that two mitochondrial oxidative stress-related genes (COX6B1 and HMGCL) were identified as diagnostic biomarkers and may serve as potential targets for IPAH development, although further research is necessary.
