Networks of pre-diagnostic circulating RNA in testicular germ cell tumour
Abstract Testicular germ cell tumour (TGCT) is a malignancy with known inherited risk factors, affecting young men. We have previously identified several hundred differentially abundant circulating RNAs in pre-diagnostic serum from TGCT cases compared to healthy controls. In this study, we performed Weighted Gene Co-expression Network Analysis (WGCNA) on mRNA and miRNA data from these samples. Central genes (hub genes) enriched functional pathways, and regulatory feature prediction were identified for all TGCT subtypes together and according to histology. The TGCT susceptibility genes TEX14 , NARS2 , and G3BP2 , were identified as hub genes in both seminoma and non-seminoma networks. We also identified UBCA1 , RCC1 , FMR1 , OAS3 , and UBE2W as hub genes associated with TGCT. The genes OAS3 and UBE2W have previously been associated with testicular dysgenesis. Furthermore, network module analysis indicated transcription factors for oestrogen-related receptors to have a potential role during development of TGCT. The overlap between mRNA network hub genes and TGCT susceptibility genes indicates a common role in TGCT development.
Evaluating historical changes in a mussel bed community in northern California
Abstract Marine foundation species are increasingly impacted by anthropogenic stressors, driving a loss of diversity within these critical habitats. Prior studies suggest that species diversity within mussel beds has declined precipitously in southern California, USA, but it is unclear whether a similar loss has occurred farther north. Here, we resurvey a mussel bed community in northern California first sampled in 1941 to evaluate changes in diversity after 78 years. More broadly, we explore the value and potential challenges of using imperfect historical data to assess community changes. Our 2019 survey documented 90 species/taxa across 10 phyla. The majority of species (~ 72%) were common to all replicate plots, suggesting that variation in species diversity over small spatial scales was unlikely to mask temporal changes. In contrast to results from southern California, we observed no decline in species diversity between timepoints. However, there were shifts in species composition, with an increase in the abundance of southern species and a decrease in northern species, consistent with warming observed at a nearby shoreline site. Overall, our findings are an encouraging sign for the health of this mussel bed community in northern California and illustrate how non-traditional data can contribute to assessments of long-term ecological change.

Fig abscission as a defense mechanism of Ficus trees against parasitism by non-pollinating fig wasps
Abstract How does the fig tree Ficus benguetensis protect its investment in the production of figs and pollinating fig wasps against parasitism from non-pollinating fig wasps? This study documents a previously overlooked defense mechanism: fig abscission—the natural shedding of the fig fruit as a defense mechanism. Our bagging experiments showed that both the absence of pollination and high parasitism levels lead to the abortion of F. benguetensis figs, with positive correlations between parasitism levels, increased abscission rates, and decreased pollinator production. Moreover, we found that high parasitism corresponds to shortened fig development periods until abscission, while medium parasitism levels result in fewer pollinators. Our findings suggest that abscission may function as a resource conservation strategy, as most of the tree’s investment in the figs occurs post-pollination. This study uncovers for the first time the use of fig abscission as a unique defense against non-pollinating fig wasp parasites, broadening our understanding of plant defense mechanisms within mutualistic interactions.

Abstract Glioblastoma multiforme (GBM) is the most prevalent, treatment-resistant, and fatal form of brain malignancy. It is characterized by genetic heterogeneity, and an infiltrative nature, and GBM treatment is highly challenging. Despite multimodal therapies, clinicians lack efficient prognostic and predictive markers. Therefore, new insights into GBM management are urgently needed to increase the chance of therapeutic success. Circulating miRNAs (miRs) are important regulators of cancer progression and are potentially useful for GBM diagnosis and treatment. This study investigated how miR-29a, miR-106a, and miR-200a affect the prognosis of GBM patients. This study was conducted on 25 GBM patients and 20 healthy volunteers as a control group. The expression levels of target miRs were analyzed pre- and post-treatment using qRT-PCR and evaluated in relation to both clinical GBM criteria and the patient’s survival modes. The diagnostic efficacy of target miRs was assessed using the receiver operating characteristic (ROC) curve. MiRs levels showed significant differences among the enrolled participants. All investigated miRs were significantly elevated in GBM patients with non-frontal lesions. Only miR-200a showed a significant difference in GBM patients older than 60 years with a tumor size ≥ 5 mm. Regarding miR-106a, a significant difference was detected based on the surgical strategy and use of an Eastern Cooperative Oncology Group (ECOG) performance status equal to 2. For miR-29a, a significant upregulation was detected according to the surgical strategy. All post-treatment miRs levels in GBM patients were significantly downregulated. In conclusion, circulating miRs revealed a significant role in predicting GBM patient treatment outcomes providing valuable insights for personalized therapeutic strategies.

Beyond despair: Leveraging ecosystem restoration for psychosocial resilience
Ecosystem restoration has historically been viewed as an ecological endeavor, but restoration possesses significant, yet largely untapped, potential as a catalyst for personal and social transformation. We highlight the opportunity for restoration to enhance community resilience by increasing agency and collective action and countering the pervasive perception that we are powerless witnesses to environmental decline. In this perspective, we take a “bright spots” approach and highlight successful examples of ecosystem restoration that have helped to nurture a sense of place, foster optimism, and cultivate stronger and more diverse social networks. These three individual- and community-level capacities have the potential to lead to increased psychosocial resilience, which is a key component of community resilience. Our aim is to spark discussion and research to better understand how we can transform restoration from a largely technical endeavor to a practice and process through which human–nature relationships are infused with deliberate meaning and human well-being is improved. With current calls to upscale and technologize restoration to meet sustainable development goals, we cannot lose sight of the value of community-engaged ecosystem restoration as a strategy with great potential for psychosocial benefits.
Assessment of salicylic acid and potassium nitrate to mitigate frost stress in autumn-sown potato crop cv. Sutlej
Abstract Potato is cultivated all the year round in Pakistan. However, the major crop is the autumn crop which is planted in mid-October and contributes 80–85% of the total production. The abrupt climate change has affected the weather patterns all over the world, resulting in the reduction of the mean air temperature in autumn by almost 1.6 °C in Pakistan, which in turn, has adversely affected the crop performance and tuber yield. This trial, therefore, was conducted to optimize and evaluate the concentration of salicylic acid (SA) and potassium nitrate (KNO3) for inducing frost stress tolerance in an autumn-sown potato crop cv. Sutlej. The findings revealed that the foliar application of 0.5 mM SA significantly enhanced the growth, yield, fluorescent, and biochemical indices of potato plants outperforming 100 mM KNO₃ application in comparison with the control. This included increased plant height by 14% and 17.6%, leaf area index by 6.3% and 26.3%, shoot biomass by 15.4% and 46.2%, crop growth rate by 16.7% and 43.3%, average tuber weight by 8.2% and 23%, tuber diameter by 6.8% and 12.2%, tuber yield by 26.1% and 46.3%, leaf angle by 16.2% and 21.6%, quantum yield of photosystem II by 20.6% and 28.2%, photosynthetically active radiations by 20.5% and 32.4%, chlorophyll content by 6.3% and 14.6%, leaf thickness by 14% and 29%, linear electron flow by 20% and 32.7%, O–2 by 6% and 14.4%, H2O2 by 11.7% and 27.6%, enzyme activities of catalase by 20.7% and 28.5%, superoxide dismutase by 28.6% and 28.5%, peroxidase by 8.3% and 13.5%, ascorbate peroxidase by 17.2% and 37.8%, total protein by 21% and 37%, proline by 36.2% and 114%, and phenolic content by 33% and 63.3% with a reduction in non-photochemical quenching by 12.7% and 29.6%, non-regulatory energy dissipation by 169.5% and 268.5%, and leaf electrolyte leakage by 57.5% and 180%, compared to KNO3 and the control, respectively. Based on the above findings, it can be concluded and recommended that 0.5 mM foliar spray of SA can be utilized on potato crop cv. Sutlej in frost-sensitive regions. However, the application rate of KNO3 needs to be optimized in order to use its maximal frost stress tolerance potential.
