The role of Big Five personality domains and facets in musical sensibility: a twin study

Abstract Musical sensibility can be understood as a propensity to be emotionally and aesthetically engaged by music and may constitute a key feature of a multidimensional definition of musicality. Yet, the nature of this construct is only just beginning to be understood. In a sample of adult Norwegian twins (N = 2592), we aimed to establish whether interindividual variability in musical sensibility may partially be attributable to personality, both in terms of the Big Five personality domains and their lower-order facets, as well as the role of genes and environments. Phenotypic analyses demonstrated that the personality domains of open-mindedness (and the facet aesthetic sensitivity in particular), agreeableness (and the facet compassion), and negative emotionality were all significantly associated with and predictive of musical sensibility. Multivariate biometric twin models further revealed that these relations were driven mainly by genetic influences, accounting for 50–100% of the observed covariance, whereas non-shared environmental influences accounted for the rest. Moreover, genetic correlations of musical sensibility with personality traits were substantial, and particularly strong for open-mindedness, pointing to considerable overlap in the biological mechanisms underlying the two traits. These findings situate musical sensibility within a broader network of psychological dispositions, possibly linked together via common affective systems.

Abstract Metabolic syndrome (MetS), marked by visceral obesity, hypertension, hyperglycemia, and dyslipidemia, elevates cardiovascular risk. Locomotive syndrome (LS), characterized by age-related mobility decline, often precedes frailty and sarcopenia. We investigated the prevalence and association between MetS and LS among Japanese middle-aged and older adults. This retrospective cross-sectional study was conducted using clinical records of 35,059 Japanese adults who underwent health check-ups between April 2021 and March 2022. MetS was diagnosed using the modified National Cholesterol Education Program Adult Treatment Panel III criteria, and LS was assessed using the LS risk test, which includes the stand-up test, two-step test, and the 25-question Geriatric Locomotive Function Scale. Among the participants, 2640 (7.5%) were diagnosed with MetS, and 5265 (15.0%) were classified as LS-positive. The prevalence of LS was significantly higher in individuals with MetS. The association between MetS and LS remained significant after adjusting for age and sex. We found that LS prevalence was higher in women than in men, though the primary association between MetS and LS remained consistent across both sexes. This large-scale real-world analysis emphasizes the need for integrating LS screening into routine check-ups to detect and manage MetS, aiding in the prevention of functional decline in aging populations.

The impact of androgen receptor pathway inhibitors as starting treatment in metastatic castration-sensitive prostate cancer on patient outcomes (OASIS Japan)

Abstract We examined the impact of starting treatment on clinical outcomes in men with metastatic castration-sensitive prostate cancer (mCSPC). This retrospective observational cohort study used claims data from the Medical Data Vision (MDV) hospital-based administrative dataset in Japan. All patients with newly diagnosed mCSPC from 1 January 2018 to 31 March 2024 were enrolled and followed up until 30 September 2024. Time-to-event analyses used Kaplan-Meier methods. The risk of death, onset of castration resistance, time to ≥ 50% PSA decline (PSA50), ≥ 90% PSA decline (PSA90), and undetectable PSA level (≤ 0.2 ng/mL) was compared between androgen receptor pathway inhibitors (ARPIs) and combined androgen blockade (CAB) or androgen-deprivation therapy (ADT) alone using a Cox proportional hazard model adjusted for age, body mass index, co-morbidities, visceral metastases, and baseline PSA. 22,559 patients with mCSPC had received relevant treatment of whom 15,797 were included in the analysis: 1167 (5.2%) started on apalutamide (APA) + ADT, 1407 (6.2%) on enzalutamide + ADT, 1262 (5.6%) on abiraterone acetate plus prednisone + ADT, and 11,961 (53.0%) on CAB/ADT alone. The median age was between 74 and 78 years in each group. Bone metastases were present in 60.5% to 72.6% of patients, visceral metastases in 2.8% to 5.7%, and nodal metastases in 19.3% to 29.4%. Overall survival and castration resistance-free survival were significantly longer in patients initially treated with APA + ADT compared to CAB/ADT (p < 0.0001 for both comparisons). In patients with regular PSA assessment, a higher percentage of patients starting with APA + ADT achieved PSA50, PSA90 and undetectable PSA at 3 months compared with CAB/ADT (p < 0.0001, p = 0.0005, p < 0.0001, respectively). Use of APA + ADT as a starting treatment for mCSPC was associated with better clinical outcomes versus traditional CAB or ADT in real-world clinical practice in Japan.

MLKL is involved in the regulation of skin wound healing and interplay between macrophages and myofibroblasts in mice

Abstract Mixed lineage kinase domain-like protein (MLKL), a critical necroptosis effector, is strongly linked to inflammation, a key component of skin wound healing. However, its precise role in the wound healing process remains inadequately characterized. This study revealed sustained MLKL overexpression throughout the wound healing process, not limited to the early inflammation phase. Wound healing was delayed in MLKL-deficient (MLKL −/− ) mice compared to wild type C57BL/6J (MLKL +/+ ) mice, with impaired morphological and pathological recovery. MLKL deficiency reduced the synthesis of inflammatory factors (IL-6, TNF-α, PGE 2 ), tissue repair molecules (EGF, VEGF, ERα, MMP-9), and apoptosis markers (Caspase-3, p53, Bcl-2) at wound site. Subsequently, a co-culture system was established to explore the roles of MLKL in macrophage-fibroblast interactions. M1 or M2 macrophages (M1ø or M2ø) were co-cultured with fibroblast-conditioned medium (MFbCM), and fibroblasts were co-cultured with macrophage-conditioned medium (M1ø CM or M2ø CM). The results indicated that MLKL +/+ M1ø CM and M2ø CM significantly increased ERα, VEGF and MMP-9 protein expression levels in fibroblasts, whereas this effect was impaired when MLKL −/− M1ø CM or M2ø CM were used. Similarly, MLKL +/+ MFbCM upregulated IL-6, NO, and TNF-α in M1ø and IL-10, arginase, and Ym-1 in M2ø, but these effects were diminished with MLKL −/− MFbCM treatment. These results indicate bidirectional crosstalk between macrophages and fibroblasts, in which MLKL plays a role. Additionally, PGE 2 was identified as a downstream mediator of MLKL-mediated interactions between macrophages and fibroblasts. In conclusion, these findings identify MLKL as a multifunctional regulator in skin wound healing in mice.
