In vitro antimicrobial and anticancer potentials of green synthesized luminescent carbon quantum dots derived from artichoke leaves

Abstract Naturally derived carbon quantum dots (CQDs) are novel carbon-based nanomaterials with excellent traits. It is highly demanded to develop CQDs from biowaste that have excellent photostability, a simple synthesis approach, and an appealing output so that they can be used widely in various fields. Herein, highly fluorescent CQDs were synthesized hydrothermally using artichoke leaves. The CQDs were synthesized and analyzed for their structure, optical properties, antimicrobial, and anticancer activities. The CQDs exhibited antimicrobial action against a single fungus strain in addition to Gram-positive and Gram-negative cells; also, the cytotoxicity against the MCF-7 cell line was evaluated as 96.5 µg/mL. The findings indicated that the spherical dots have a semi-spherical shape with the smallest size of 2.88 nm, and a zeta potential value of 37.31 V, thus confirming that the synthetic CQDs are in an outstanding colloidal state. When photoexcited at 320 nm, the dots were found to show blue fluorescence at 398 nm with a fluorescence quantum yield of 3.32%, long fluorescence decay time, high photostability, and good sensing for hydrogen peroxide. Additionally, the effect of ionic strength was evaluated.

A cytidine deaminase-like protein modulates pyrimidine nucleotide homeostasis in Trypanosoma brucei

Abstract Pyrimidine nucleotide homeostasis is critical for DNA replication and cell viability, yet its regulation in Trypanosoma brucei , the causative agent of African trypanosomiasis, remains poorly understood. Here, we characterize a T. brucei cytidine deaminase-like protein (TbCDA-like), a kinetoplastid-specific enzyme absent in mammals, that harbors a deaminase domain and a zinc-finger CCCH motif. Using RNA interference (RNAi) and overexpression approaches, we demonstrate that TbCDA-like modulates pyrimidine nucleotide pools, influencing both ribonucleotide and deoxyribonucleotide profiles. Overexpression of TbCDA-like resulted in a substantial reduction of dCTP and CTP levels while elevating dTTP and UTP pools, suggesting a role in cytidine derivative deamination. Conversely, RNAi-mediated depletion of TbCDA-like caused CMP accumulation and reduced dTTP levels. Notably, overexpression induced severe cytotoxicity, growth arrest, DNA damage and cell cycle defects, evidenced by sub-G1 populations, increased nuclear H2A phosphorylation and aberrant kinetoplast and nuclear morphologies. Localization studies revealed that TbCDA-like is primarily cytosolic and relocalizes to stress granules upon nutrient deprivation, suggesting a role in RNA metabolism. These findings establish TbCDA-like as a key regulator of pyrimidine nucleotide homeostasis in T. brucei , linking nucleotide imbalances to DNA replication stress and genome instability. Given its absence in mammals, TbCDA-like presents an attractive target for therapeutic intervention against trypanosomiasis.

Attention to facial emotions in adult women varies by type and severity of childhood maltreatment experience and emotion regulation strategy

Abstract Little eye-tracking research has been conducted so far on the association of subtypes of childhood maltreatment and habitual emotion regulation with attention to emotions. To address this issue, gaze behavior of one hundred women with experiences of childhood maltreatment was examined in a free-viewing task in which pairs of faces consisting of an emotional (happy, surprised, angry, disgusted, fearful, and sad) and a neutral face were displayed. Participants’ childhood maltreatment experiences, emotion regulation strategies (reappraisal and suppression), anxiety, depression, stress perception, level of alexithymia, and intelligence were assessed. Fixation durations were used to assess attentional bias. A mixed linear model with post-hoc pairwise comparisons was used to investigate research questions. Results indicated that, at high levels of maltreatment severity, attentional preference for positive over other emotional faces was found to be diminished for physical and emotional abuse but not for emotional neglect. At a severe level of emotional abuse, a bias away from disgusted faces was observed. Suppression was linked to reduced and reappraisal was related to heightened attention allocation to facial emotions. Our data indicates that attention to social emotional information in adulthood could depend on the type and severity of childhood trauma and habitual emotion regulation strategy.

Abstract When planning an action sequence, it has been shown that humans prune decision trees to reduce computational complexity, instead of considering all possible options. However, little is understood about pruning employed in probabilistic environments, where actions result in multiple outcomes with varying probabilities, and how decision biases, such as discounting of probabilistic rewards, influence decisions. This study investigates whether participants prune low-probability options in a three-step decision-making task and analyzes the impact of probability discounting on planning. Potential age-related differences in planning strategies are explored in groups of young (aged 18–35 years; n = 57) and older (aged 65–75 years; n = 50) adults. By using reinforcement-learning modeling and model comparison, we show that participants reduce computational demands by pruning decision tree branches of lower probability—a highly efficient strategy in this environment. Additionally, participants reduce their planning depth, i.e., the number of considered steps. Planning is further influenced by discounting high-probability outcomes. Older individuals show stronger reductions in planning depth, an increase in decision noise, and more pronounced probability discounting, which contributes to the observed age-related decline in planning performance. Our findings suggest directions for future research to elucidate the underlying meta-control mechanisms guiding the application of planning strategies.

Multi-channel volume density neural radiance field for hyperspectral imaging

Abstract Hyperspectral imaging and Neural Radiance Field (NeRF) can be combined in powerful ways. With limited hyperspectral images, NeRF can generate images of objects with spectral information from arbitrary viewpoints, which can effectively mitigate defects such as long acquisition time and difficulty in obtaining hyperspectral images. This paper addresses challenges in the application of NeRF methods in the hyperspectral domain, including local errors in convergence caused by noise. Leveraging the characteristics of hyperspectral data, we propose a neural radiance field method employing a multi-channel volume density distribution function. This approach alleviates issues during the generation of neural radiance fields from hyperspectral data, enhancing the robustness of hyperspectral neural radiance field methods across various scenarios, which can help downstream tasks such as discriminating objects more effectively than RGB methods. Experiments demonstrate that the proposed method generates superior hyperspectral images under diverse conditions, with a maximum PSNR 37.66 and a maximum SSIM 0.982.

Reexamination of honey bee Africanization in Mexico and other regions of the New World

Abstract Honey bees (Apis mellifera) are not native to the New World. The initial introduction of the species to the Americas occurred from Europe, with subsequent introductions from Africa. The African bees hybridized with European bees and are now referred to as Africanized bees. A large feral population was established and subsequently colonized extensive areas of both the North and South American continents, including Mexico. The aim of this study was to conduct a morphometric analysis of geographic variation among Africanized bees. Recently acquired data from Southeastern Mexico were compared with existing datasets of Africanized bees and evolutionary lineages from the Old World. The forewing venation was described using 19 landmarks. The honey bees originating from southeastern Mexico exhibited significant differences from all other investigated populations. It is necessary to verify if the observed geographic variation within Africanized bees is related to natural selection or other factors, including hybridization or genetic drift. Furthermore, honey bees from populations in the USA and Argentina, which appear to have not been affected by Africanization, differed markedly from honey bees naturally occurring in Europe and can be classified as hybrids between evolutionary lineages.
