Fast detection of synergy and antagonism in antifungal combinations used against Candida albicans clinical isolates
Abstract The rise in antifungal resistance has limited treatment options for serious fungal infections, emphasizing the need for effective combination therapies. However, low-cost and rapid systems to evaluate synergy and antagonism in antifungal combinations are lacking. Here, we introduce a novel in vitro testing method for assessing antifungal interactions in C. albicans, enabling the simultaneous testing of three antifungal agents in a single agar plate with overnight results. This method, validated against the checkerboard assay, provides consistent fractional inhibitory concentration (FICi) measurements with reduced variability and workload. We applied this method in a comprehensive screen of 92 clinical C. albicans isolates for three antifungals—amphotericin B, fluconazole, and anidulafungin—yielding assessments of a total of 276 distinct combinations of antifungals and isolates. Results revealed isolate-specific interaction patterns, with amphotericin B and fluconazole showing synergy in 1% of isolates, anidulafungin and fluconazole in 19.5%, and amphotericin B and anidulafungin in 23.9%. These findings underscore the need for isolate-specific testing in clinical settings. This proposed assay aims to present a solution to that as a scalable high throughput approach to this clinical problem.

Effect of mesh sizes on the mechanical properties of stainless-steel wire mesh/glass fiber reinforced hybrid composite laminates for aerospace applications
Abstract This study investigates the effect of stainless-steel wire mesh (SSWM) size on the mechanical performance of glass fiber–SSWM hybrid composites for aerospace application. Laminates were fabricated by embedding AISI 304 steel meshes (10 to 120 openings per inch) and bi-directional glass fibers in epoxy using hand layup and compression molding. Mechanical tests showed mesh size strongly affects properties: Mesh 120 achieved the highest tensile strength (539.19 MPa) with low void content (0.49%), suitable for tensile-critical parts; Mesh 20 had the highest flexural strength (487.97 MPa) due to its heavier mesh weight and low voids (0.53%), ideal for load-bearing panels; Mesh 40 offered the best balance of impact resistance (1.715 J), tensile (409.10 MPa), and flexural (339.78 MPa) strengths, supporting multifunctional structures. Finer meshes (80, 120) exhibited improved ductility and damping for vibration-sensitive applications. SEM analysis revealed strong interfacial bonding and crack-bridging in Mesh 40, while Mesh 120 showed microcracking and fiber pull-out, indicating trade-offs between tensile and impact performance. Void content ranged from 0.49% to 1.93%, confirming high manufacturing quality. These findings emphasize the critical role of mesh geometry in optimizing hybrid composite performance and suggest further work on advanced fabrication methods and durability testing.

Care professionals’ accounts of providing support and treatment for people with co-occurring alcohol use disorder and depression in the North East of England, UK: A qualitative study informed by complexity theory
Introduction There is an acknowledged care gap for patients with co-occurring substance use and mental ill-health. This study sought to use complexity theory to help make sense of the experiences of people who deliver or commission formal care for patients with alcohol use disorder and depression across one specific health and social care system. Method Qualitative interviews with 26 health and social care professionals in the North East and North Cumbria Integrated Care System, England, were conducted. Data analysis was undertaken using reflexive thematic analysis and informed by key concepts from complexity theory. Results Three main themes were identified: (1) how the interplay between risk, stigma and resource pressures influences how care professionals interpret and apply practice guidelines; (2) how individualised and disjointed practices have structural and historical roots, in particular the impact of health service commissioning cycles; (3) ways in which practitioners have been able to adapt and engage in creative practice to temporarily plug gaps in care. Conclusions The pressure of working with increasingly scarce resources, within a highly fragmented, shifting, and risk-averse care infrastructure, adversely affected professionals’ capacity to provide consistent, patient-centered support. Innovations have emerged that address some of these barriers, but further investment is needed to better support the substance use and mental health workforce, including lived experience peer workers.
