Abstract The Li-mediated nitrogen reduction reaction (Li-NRR) has been proposed as one of the most promising ambient production routes for green ammonia. However, the effect of the applied potential ( E we ) on the reaction performance and the properties of the solid electrolyte interphase (SEI) remain poorly understood. Herein, we combine potential controlled experiments using a reliable Li x FePO 4 based reference electrode with post-mortem SEI characterization techniques, wherein we observe both an increase in the LiF concentration in the SEI, originating from LiTFSI decomposition, and the Faradaic efficiency (FE NH3 ) with an increasing E we . The transition from a predominantly organic SEI at low E we (−3.2 V SHE ) to a LiF-enriched layer at higher E we indicates the existence of kinetic barriers for the SEI formation reactions. Moreover, thicker and denser SEI structures observed at a higher E we enhance the Li-NRR by improving the mass transport regulation between reactant species. However, these thicker and denser SEI morphologies lead to current instabilities due to dynamic SEI thickening and breakdown.

Roles of histone chaperone Nap1 and histone acetylation in regulating phase-separation of nucleosome arrays

Abstract Chromatin condensation is dynamically regulated throughout the cell cycle and plays key roles in modulating gene accessibility. The DNA-histone dynamics in the nucleosome are central to the regulation mechanisms of chromatin condensation, which remain poorly understood. Employing fluorescence recovery after photobleaching, optical super-resolution imaging, and microrheology with optical tweezers, we investigated the roles of various parameters in regulating phase-separation of 12-mer nucleosome arrays. Here, we show that histone H4 tail lysine residues are the main drivers of liquid-liquid phase separation of nucleosome arrays. We also show that the condensed liquid-like droplets comprise a mobile fraction and a relatively immobile structural scaffold. Histone chaperone Nap1 and histone H3 tail acetylation enhance DNA-histone dynamics within this scaffold, thereby lowering the overall viscosity of the droplets. These results suggest that histone chaperone and histone H3/H4 tails play critical roles in regulating chromatin condensation and gene accessibility in condensed chromatin.

Pruned U-net with multi-scale feature fusion and attention for real-time UAV remote sensing of levee defects

Abstract The long-term performance of levee infrastructure is increasingly threatened by environmental exposure and material degradation, underscoring the need for efficient, accurate inspection. Unmanned Aerial Vehicle (UAV)-based remote sensing offers a cost-effective solution, enabling rapid acquisition of high-resolution imagery over large surfaces; however, stains, occlusions, and illumination variability frequently degrade automated detection. To address these challenges, we propose a real-time semantic segmentation framework built on an optimized U-Net. The model integrates structured pruning to accelerate inference, a residual convolutional block attention module (ResCBAM) to suppress background interference and enhance defect saliency, and a multi-scale feature-fusion strategy with online feature distillation to strengthen fine-grained representations across resolutions. We evaluate the approach on UAV imagery collected from an aged levee section. The proposed method attains 90.05% accuracy, 88.94% recall, 89.22% precision, and 88.67% IoU, outperforming state-of-the-art baselines, while achieving a real-time processing rate of 57.74 FPS. These results demonstrate that the framework delivers a favorable speed–accuracy trade-off and is suitable for large-scale UAV-based levee monitoring. Overall, the experiments indicate strong potential for timely defect identification and proactive risk management in levee systems.

Compressed sensing expands the multiplexity of imaging mass cytometry

Abstract The multiplexity of current antibody-based imaging is limited by the number of reporters that can be detected simultaneously. Compressed sensing can be used to reconstruct high-dimensional information from low-dimensional measurements. Previously, compressed sensing using composite in situ imaging (CISI) of transcriptomic data leveraged gene co-regulation structure to recover spatial expression of 37 RNA species from images of 11 composite channels. Here, we extend the CISI framework to protein expression data measured by imaging mass cytometry (IMC). CISI-IMC accurately recovers spatial expression of 16 immune and stromal marker proteins from images of 8 composite channels with an average Pearson’s correlation of 0.8 across protein. Training the CISI-IMC framework using data collected on multiple human tissues enables universal decompression of composite data from a wide range of tumor and healthy tissue types. The expression dictionary and barcoding matrix described here are immediately implementable for general immune and stromal cell type classification, but CISI-IMC can in principle be applied to other markers or other antibody-based imaging methods. Our work lays the foundation for much higher plex protein imaging.
