Abstract Ewing sarcoma (EWS), a rare pediatric bone tumor, poses unique therapeutic challenges due to its distinct microenvironment and limited molecular understanding. To gain a comprehensive molecular and functional view of the tumors in their microenvironment, we perform a deep mass spectrometry-based proteomic analysis of 170 tumor samples from 74 patients from primary, relapsed, and metastatic tumors. Analysis of more than 10,000 proteins across patients reveals insights into cancer prognosis, chemo-resistance, and progression. Our analyses suggest that ferroptosis pathways may be associated with chemotherapy response in EWS, and we delineate molecular subclasses that correlate the tumor immune landscape with DNA damage repair, ubiquitin-related proteins, and patient outcomes. Multiplexed immunofluorescence imaging indicates possible associations between neutrophils and poorer prognosis, and between macrophages/T cells and a more favorable prognosis. Altogether, this investigation provides valuable insights into the intricate biology of EWS, paving the way for developing therapeutic strategies.

Abstract Wireless bioelectronics demand transduction strategies that are simultaneously sensitive, noise-resilient, and biologically safe. Conventional wireless sensors typically rely on dielectric capacitors with inherently low capacitance, necessitating operation at MHz frequencies. Such high-frequency coupling often introduces electromagnetic interference, tissue heating, and degraded signal fidelity in biological environments. Here we present a wireless low-frequency electrochemical sensing (WiLECS) platform that couples ionic dynamics with low-frequency LC resonant circuits. The device combines a biocompatible ion gel, composed of a choline-malate ionic liquid embedded in a chitosan matrix with functionalized Au nanoparticles, with a miniaturized LC antenna. Unlike conventional capacitive sensors, WiLECS employs piezo-driven ion redistribution to modulate the dielectric environment of the circuit, enabling sustainable wireless transduction below 1 MHz with high sensitivity and reliability. This approach directly bridges ionic dynamics and electronic resonance, allowing mechanical stimuli to be transduced into biologically safe low-frequency electronic signals. As proof of concept, we demonstrate real-time wireless blood-pressure monitoring in artificial arteries with atherosclerotic plaque, showing resolution of subtle pressure variations under clinically relevant conditions.

Phytochemical profile and chemosensitizing anticancer activity of Mitragyna speciosa and mitragynine

Abstract This study examined the phytochemical composition, antioxidant efficacy, and cytotoxic properties of Mitragyna speciosa (kratom) extracts and its principal alkaloid, mitragynine. The extracts exhibited significant antioxidant properties, and chemical analysis verified mitragynine as the principal alkaloid, aligning with the distinctive phytochemical profile of M. speciosa. In vitro cytotoxicity studies demonstrated that both kratom extract and mitragynine suppressed cancer cell proliferation (A549, KKU213C, and HeLa cancer cell lines) in a concentration- and time-dependent manner, with mitragynine displaying greater potency compared to the crude extract, especially against cholangiocarcinoma cells. Importantly, synergistic cytotoxic effects were noted when kratom extract or mitragynine was combined with standard chemotherapeutic agents across various cancer cell lines, especially HeLa cervical cancer. These effects correlated with diminished expression of the anti-apoptotic protein BCL-2. In general, the results show that M. speciosa extract and mitragynine are chemosensitizers that could make standard chemotherapy work better. Additional research is necessary to clarify the molecular mechanisms involved and to confirm these effects in vivo.

Abstract The regular inspection of the eye health is an essential part of the veterinary care task in an animal facility in order to achieve robust results during experimentation. The aim of the study was to find out, when and how ocular diseases are recorded in laboratory animal facilities and what measures are taken. An online questionnaire was sent to people who work in a research environment and are involved in monitoring of animal wellbeing. In addition, an evaluation of 142 veterinary clinical cases in mice were carried out under routine conditions with the help of ophthalmological tools. 128 participants from three European countries took part in the survey. 93% stated, that eye changes, if recognized, are recorded during cage changing but in 70% of the responses, there was no systematic analysis of the underlying causes. The population study showed that technical aids are indispensable for the detection of eye lesions in mice. The survey and the population study show that although recommended, routine checks of the eye are rarely carried out and usually without suitable technical aids. From the point of view of good veterinary practice, more attention should be paid to the ocular health in laboratory animal husbandry.
