Abstract

e16375 Background: Pancreatic cancer is a highly aggressive malignancy with an extremely poor prognosis, often referred to as the "king of cancers." Its five-year survival rate is less than 10% due to late diagnosis, rapid progression, and resistance to therapy. Gemcitabine, a first-line chemotherapy drug for pancreatic cancer, has shown some efficacy in extending survival. However, its therapeutic benefits are often limited by the rapid development of drug resistance. These factors collectively pose significant challenges for effective treatment and highlight the urgent need for novel therapeutic strategies. Methods: Elucidating the novel molecular mechanisms of gemcitabine resistance in pancreatic cancer using molecular biology, cell biology, and other approaches. Results: We found that the DEPTOR protein directly regulates the glutamine metabolism pathway to promote drug resistance in pancreatic cancer. Conclusions: "DEPTOR protein can serve as a molecular marker for gemcitabine resistance in pancreatic cancer."
