Abstract

Abstract Human intestinal Organoids (HIOs) may be set up from primary human tissue following dissociation, release, and culture of intestinal LGR5 + stem cells. Here, we characterise the use of a semi-automated mechanical tissue dissociation technique, which demonstrates an expedited workflow and streamlined protocol, standardising key aspects of the process while consistently maintaining a high level of cultured organoid viability.
